The Serbian took three days to defeat Spain's Roberto Bautista Agut 3-6, 6-4, 6-1, 7-5 in their fourth round rain-interrupted contest.
Djokovic, who had US$99,673,404 at the start of the tournament, achieved the landmark figure after moving into a 28th consecutive Grand Slam quarter-final.
He will next face Czech seventh seed Tomas Berdych for a place in the semi-finals.
Last year, Djokovic's hopes of completing a career Grand Slam were crushed after he lost to Stan Wawrinka in the pulsating finale of the French Open.
Seventeen-time Grand Slam champion Roger Federer stands second on the all-time list with US$98,011,727 career prize money.
