In Hum Tum, Saif Ali Khan plays Karan Kapoor, a renowned cartoonist. He creates a cartoon strip called Hum Tum because he sees the world divided into two parts: male and female.

When he meets Rhea Prakash (Rani Mukerji) for the first time in Amsterdam, the two start off on the wrong note. To ease the situation, Karan kisses her. It only worsens things.

Six months later, the two meet again, this time in New York. Still ruffled about the kiss, Rhea spills the beans to Karan's then girlfriend, who promptly dumps him.

Soon, Karan and Rhea become friends. Whenever she goes through emotional tragedies, Karan helps her out. He even tries to fix her with a friend of his.

Ten years pass.

Their friendship stands the test of time. Yet, Karan always manages to get on the wrong side of Rhea. Will the duo set aside their differences?
